Everton shared a good tip in the comments below. What he had done is change the name of the comments submission page, so that spam robots that are trying to automatically leave comments on the default page (wp-comments-post.php) can’t as it no longer exists or has that function.

Simple Trackback Validator
I’ve installed this plugin in addition to Spam Karma to reduce the amount of trackback spam I have to bin daily. What the Simple Trackback Validator plugin does is automatically reject any trackback that doesn’t actually link to the article that it is pointing to (SK2 will only flag these trackbacks as spam, but not auto-delete). This is such a simple step, but it has significantly reduced the amount of spam in my WordPress inbox.
Math Comment Spam Protection
I was still getting more spams from SK2 daily than I wanted so I also installed the Math Comment Spam Plugin. I’ve set this up so any visitor who isn’t logged in has to answer a simple math question before they can leave a comment. This is intended to prove that the visitor is a human being and not a spam robot, and has done a very good job of reducing comment spam.
I did try Bad Behaviour which stops users it thinks are spammers even accessing my site. However, after being told by a few friends that it was incorrectly stopping them accessing the site I decided to turn it off.
